ABSTRACT

This study investigated the mechanism of Danggui Shaoyao Powder(DSP) against mitophagy in rat model of Alzheimer's disease(AD) induced by streptozotocin(STZ) based on PTEN induced putative kinase 1(PINK1)-Parkin signaling pathway. The AD rat model was established by injecting STZ into the lateral ventricle, and the rats were divided into normal group, model group, DSP low-dose group(12 g·kg~(-1)·d~(-1)), DSP medium-dose group(24 g·kg~(-1)·d~(-1)), and DSP high-dose group(36 g·kg~(-1)·d~(-1)). Morris water maze test was used to detect the learning and memory function of the rats, and transmission electron microscopy and immunofluorescence were employed to detect mitophagy. The protein expression levels of PINK1, Parkin, LC3BⅠ/LC3BⅡ, and p62 were assayed by Western blot. Compared with the normal group, the model group showed a significant decrease in the learning and memory function(P<0.01), reduced protein expression of PINK1 and Parkin(P<0.05), increased protein expression of LC3BⅠ/LC3BⅡ and p62(P<0.05), and decreased occurrence of mitophagy(P<0.01). Compared with the model group, the DSP medium-and high-dose groups notably improved the learning and memory ability of AD rats, which mainly manifested as shortened escape latency, leng-thened time in target quadrants and elevated number of crossing the platform(P<0.05 or P<0.01), remarkably activated mitophagy(P<0.05), up-regulated the protein expression of PINK1 and Parkin, and down-regulated the protein expression of LC3BⅠ/LC3BⅡ and p62(P<0.05 or P<0.01). These results demonstrated that DSP might promote mitophagy mediated by PINK1-Parkin pathway to remove damaged mitochondria and improve mitochondrial function, thereby exerting a neuroprotective effect.

ABSTRACT

Objective To establish the plasma fingerprint of Danggui Shaoyao Powder (DSP) for the analysis of the transitional components in rat plasma after administration of DSP extracts Methods Eleven batches of rat plasma were prepared after oral administration and the plasma fingerprint was established by UPLC-UV. The transitional components in rat plasma after administration of DSP extracts was analyzed by UPLC-Q/TOF-MS. Results The plasma fingerprints of common peaks for 11 batches of DSP were established to ascertain the optimized blood collection time, and the method was used to process the plasma. Fifteen common peaks were detected in plasma fingerprint, and the similarity were both higher than 0.933. The methodology of plasma pharmacochemistry was adopted to analyze the common peaks, and 15 transitional components, including 10 prototype components and five metabolites were identified. Conclusion The established plasma fingerprint of DSP provide the basis for the further study of transitional components.

ABSTRACT

Objective To investigate the curative effect of modified Danggui Shaoyao Powder in the treatment of patients with chloasma and the effect on the expression of cyclooxygenase-2 (COX-2) and nuclear factor E2 (Nrf2) in serum.Methods90 cases of patients with chloasma treated in the hospital were selected as the subjects.According to different treatment methods, the patients were divided into the control group (treated with routine western medicine, n=48) and the study group (treated with modified Danggui Shaoyao Powder, n=42).The clinical efficacy, changes of cytokines (serum COX-2 and Nrf2), patch pigment regression.ResultsAfter treatment, the total effective rate of the study group was significantly higher than that of the control group (90.48% vs 72.92%) (P<0.05).After treatment, COX-2, patch pigment regression scores in the two groups were significantly lower than those before treatment, and Nrf2 was significantly higher than that before treatment.The changes were more significant in the observation group than the control group (P<0.05).ConclusionModified Danggui Shaoyao Powder is significant effective in the treatment of chloasma.It can effectively promote the regression of pigment, reduce the skin lesion area and improve the quality of life of patients.The mechanisms may be related to effectively reducing COX-2 and increasing Nrf2.

ABSTRACT

OBJECTIVE:To investigate the effect of Danggui shaoyao powder (DSS) on uterine structure and expressions of estrogen receptorα(ERα),estrogen receptorβ(ERβ)in uterine cavity epithelium and matrix in model rats in perimenopausal peri-od. METHODS:40 female SD rats were randomly divided into sham operation group (normal saline),model group (normal sa-line),DSS low-dose,medium-dose,high-dose groups(1.94,3.87,7.44 g/kg),8 in each group. Except that rats in sham opera-tion group received resection of fat nearby ovaries,rats in other groups received resection of bilateral ovaries to induce models in perimenopausal period. After modeling,rats were intragastrically administrated once a day,for 8 weeks. After administration,wet mass of uterine was weighted. Changes in uterine morphology and structure of rats were observed,expressions levels of ERα and ERβ in uterine cavity epithelium and matrix were determined. RESULTS:Compared with sham operation group,rats in model group showed low columnar in endometrial columnar epithelium,lamina propria layer,muscular layer and serous layer were signifi-cantly atrophied,stromal cells had obvious nuclear condensation. There was marked decrease in uterine wet mass,uterine cavity ar-ea and endometrial thickness as well as the number of uterine glands(P<0.01),and the expression levels of ERαand ERβin uter-ine cavity epithelium and matrix were significantly reduced(P<0.01). Compared with model group,the atrophy degree of endome-trium and lamina propria layer had no significant differences in DSS each dose groups. However,lamina propria layer was rich in glands,and there were significant differences in uterine wet mass,uterine cavity area,endometrial thickness,ERα expression level in uterine cavity epithelium and matrix (P>0.05). However, the number of uterine glands in DSS medium-dose,high-dose groups was significantly increased(P<0.01),and ERβexpres-sion level in uterine cavity epithelium and matrix in DSS high-dose group was significantly increased (P<0.05 or P<0.01). CONCLUSIONS:DSS has not obvious effect on im-proving the symptoms of uterine gland atrophy of model rats in perimenopausal period,but it can increase the number ofuterine glands,and the mechanism may be associated with improving the ERβ expression level in uterine cavity epithelium and ma-trix.

ABSTRACT

OBJECTIVE:To establish a method for the content determination of peoniflorin in Danggui shaoyao powder,and provide a reference for controlling the quality of the preparation. METHODS:HPLC was performed on the column of Symmetry C18 with mobile phase of acetonitrile-water(containing 0.1% phosphoric acid)(14∶86,V/V)at a flow rate of 1.0 ml/min,detection wavelength was 230 nm,column temperature was 20℃,and injection volume was 20μl. RESULTS:The linear range of peoniflo-rin was 10-80 μg/ml(r=0.999 3);RSDs of precision,stability and reproducibility tests were lower than 2%;recovery was 98.3%-104.9%(RSD=2.0%,n=9). CONCLUSIONS:The method is simple,accurate and specific,and can be used for the con-tent determination of peoniflorin in Danggui shaoyao powder.

ABSTRACT

Objective To explore the effect of Danggui shaoyao powder combined with Sanhuang lotion fumigation on serum TGF-β1,IL-4 and IL-10 in the treatment of chronic pelvic inflammatory disease.Methods 38 cases with chronic pelvic inflammatory disease were selected and divided into the control group (19 cases) and the experiment group(19 cases) according to the random number table.The control group were treated with Sanhuang lotion fumigation bath, the experiment group were treated on the basis of the control group with Danggui shaoyao powder, 10 days for a course of treatment, a total of two courses of treatment.The serum IL-4, IL-10, TGF-β1 and clinical efficacy were compared before and after treatmentbetweentwo groups.Results The total effective rate of the experiment group was 94.74%, which was higher than 84.21% of the control group(P<0.05).After treatment, the clinical symptom score in experiment group was lower than that in control group, the serum levels of IL-4 and TGF-β1 were lower than those in control group, the serum level of IL-10 was higher than that in control group (P<0.05).Conclusion Danggui shaoyao powder combined with Sanhuang lotion fumigation could decreases IL-4 and TGF-β1 levels, increase IL-10 level and improve the inflammatory state in the treatment of chronic pelvic inflammatory disease.

METHODS: Neovascular glaucoma patients were enrolled from January 2011 to February 2014 in our hospital, according to the different methods of treatment were divided into observation group given traditional Chinese medicine and Western medicine combined with trabeculectomy, and control group received Western medicine combined with trabeculectomy. ABSTRACT

Objective: To explore the effects of Danggui Shaoyao Powder (DSP) on in vivo uterine contractions and vasomotor factors of rats, and to investigate the mechanism of DSP for healing primary dysmenorrhea. Methods: Female SD rats were randomly divided into model, Verapamil (100 μg/kg, positive control), low-, mid-, and high-dose DSP (4, 8, and 16 g/kg) groups, and the rats were administered once daily for 7 d. The rats were sc administered with diethylstilbestrol (4 mg/kg) on day 7, on the next day the in vivo uterine contraction of rats was observed before and after oxytocin treatment under anesthesia condition, and the levels of nitric oxide (NO) and endothelin (ET) in uterine homogenate were determined. Results: DSP increased the average muscle tension of uterine contraction of normal rats obverously, but inhibited the oxytocin-induced uterine contraction significantly. The changes of the uterine contraction frequency, amplitude, average muscle tension, and uterine vigour of rats before and after oxytocin processing were significantly inhibited. DSP increased the NO level in the uterine of the experimental animals after oxytocin treatment and reduced the level of ET. The NO/ET ratio increased significantly. Conclusion: DSP could inhibit the in vivo uterine contraction, regulate the vasomotor substances, and improve the uterine blood supplement, resulting in the beneficial effect on treating primary dysmenorrhea.

ABSTRACT

AIM: To isolate,identify and screen active compounds of simplitied formula of Danggui Shaoyao Power(poria,Rhizoma Atracty-lodis Macrocephalae,and Radix Angeliae Sinensis). METHODS: Compounds in the fraction of CO_2 supercritical fluid extraction(SFE-CO_2) were isolated by silica gel chromatography and detected the protective effect on KCl-induced PC12 cell damage. RESULTS: Twelve compounds were obtained,and nine of them were identified as atractylenolide Ⅰ,Ⅱ,Ⅲ,biatractylenolide,levistolid A,pachymic acid,ferulic acid,?-sitosterol and ?-daucosterol,of which only atractylenolide Ⅲ can improve the cell viability significantly. CONCLUSION: Atractylenolide Ⅲ may be one of the active compounds of FBD in curing vascular dementia.
